国立精神・神経医療研セと生理研、JST、手の把握動作に関わる新たな神経機構を発見  from 日経バイオテクONLINE  (2013-5-15 6:00)

国立精神・神経医療研究センター神経研究所モデル動物開発研究部の武井智彦室長と関和彦部長らは、手先で器用に物を扱う把握動作の際に活動する新たな神経機構を解明した。2013年5月15日（米東部時刻）に発行される米神経科学学会のThe Journal of Neuroscience誌で発表する。この成果のプレスリリースは、関部長に戦略的創造研究推進事業個人研究（さきがけ）の研究費を助成している科学技術振興機構（JST）と、関部長と武井室長が09年9月まで在籍した自然科学研究機構生理学研究所が、国立精神・神経医療研究センターと共に発表の主体となった。

東京大学医科学研究所幹細胞治療研究センターの中内啓光教授と山崎聡助教らは、長期にわたって造血能を保持できる機能的な造血幹細胞を、iPS細胞から作製する方法を開発した。iPS細胞をマウスに注入するとできる奇形腫テラトーマの中で造血幹細胞が分化し、骨髄に移動することを見いだした。造血幹細胞の維持に必要とされるサイトカインであるSCFとTPOを持続補給できるサイトカインポンプと、ストローマ細胞であるOP9細胞とを、iPS細胞と同時にマウスに移植することにより、テラトーマの中で造血幹細胞が誘導される効率を向上させた。マウスiPS細胞に続いてヒトiPS細胞からも造血幹細胞を作製できた。さらに、共通γ鎖（γc）遺伝子を欠損しているためにリンパ球が無く重症免疫不全症（X-SCID）を発症するマウス（X-SCIDマウス）からiPS細胞を作り出し、正常なγc遺伝子を導入したiPS細胞をOP9細胞と共にX-SCIDマウスに移植したところ、免疫不全を克服して生存できることを見いだした。この成果は、米遺伝子治療学会のMolecular Therapy誌（インパクトファクター6.873、Nature Publishing Groupが発行）のオンライン版で2013年5月14日（米東部時刻）に発表する。中内教授は ...
